Description:
The overall objective of this Project is to develop the technology and expertise necessary for reprogramming the human genome through targeted integration of recombinant DNA within induced-pluripotent stem cells (iPSCs). This Project aims to leverage the innate programming of the human genome by performing integration of therapeutic genes, such as chimeric antigen receptors (CAR), into targeted sites. These tools will be key to the deployment and validation of NRC’s self-cleaving and integrating plasmid (pSCIP) technology towards engineering iPSC-derived CAR-Natural Killer (NK) cells that will prevent treatment-associated toxicity and immune-mediated adverse events and help expedite preclinical proof of concept studies for targeting solid tumors such as triple negative breast cancer.
