Description:
The project aims to further develop the D19 clone into a user-friendly cell line for producing AAV vectors, enhancing supply for laboratories working on AAV-based gene therapies. The D19 cell line was generated by NRC from suspension and serum-free media-adapted HEK293SF cells. Using lentiviral vectors, this parental cell line was modified to integrate into its genome a gene expression regulation system induced by the addition of cumate and coumermycin to express AAV Rep proteins (Rep40 and Rep68). Three of the best clones produced AAV titers comparable to those obtained by standard triple plasmid transfection of their parental cells. Anticipated outcomes include expanding AAV production capacity, enhancing market position, generating revenue for further technology development, achieving a breakthrough in AAV bioproduction by reducing costs and streamlining industrial scaling, and strengthening Canada’s biomanufacturing capabilities in gene therapies to enhance autonomy in high-value pharmaceuticals
